The unmanned freighter took off from a spaceport in the US state of Virginia on a Northrop Grumman Antares-230 rocket, NASA tweeted late on Wednesday.
Cygnus is expected to reach the space station on Friday. Using a robotic arm, US astronaut Anne McClain and her Canadian colleague David Saint-Jacques will help the spacecraft dock.
Cygnus is carrying material for scientific experiments and food packs.
