Sergeant Barry Hills said the signs, from Coomoora to Malmsbury, had been run over by a car on Saturday night and would now have to be replaced.
"They are completely flattened so we believe it might have been a bull bar which was used to do it," he said.
"It's certainly unusual, especially because it has occurred on both sides of the road."
He said the cost, believed to be upwards of a thousand dollars, would fall on VicRoads.
"We are appealing to anyone who may have seen a vehicle, or anyone acting strange on Saturday night or Sunday morning to contact us," he said.
Witnesses can call Daylesford Police on 5348 2342 or Crimestoppers on 1800 333 000.
