Cr Klein said the decision, which was not an easy one to make, was due in part to his starting a new job in Melbourne.
Cr Klein lost his license for six months in June and has been reliant on public transport to attend events.
Councillors have two years left of their terms, meaning a by-election has been triggered.
Cr Klein served as mayor for the 2011-12 and 2016-17 terms.
