The nine-kilometre track links the historic old Trentham Racecourse with a number of existing reserves close to the township.
An enthusiastic crowd of more than sixty walkers and cyclists joined to celebrate the milestone.
Hepburn Shire councillor Sebastian Klein said the community had been persistent in getting the project off the ground.
He said the track showcased some of the hidden gems of Trentham, which was a perfect town for walking.
"People wont appreciate a thing until they know it," he said.
"This is about the community introducing their environment to other residents and the state.
"Active participation is also a catch-cry of preventative health and this is all part of that."
In 2010, the group secured funding from Hepburn Shire and the state government.
The trail starts and finishes at the Quarry Street reserve, providing access to unspoilt parts of the Wombat State forest.
Works included a complete regrade and refurbishment of the Old Trentham Racecourse track, spraying and removal of gorse, broome and blackberry, and repairs to the existing four-wheel-drive access tracks so they are accessible to walkers and cyclists.
Details of the route will be available at each of the information centres within the shire.
